Oveporexton Trials in Narcolepsy Type 1

About this episode

Two phase 3 clinical trials evaluated the efficacy and safety of oveporexton, an oral orexin receptor 2-selective agonist, for treating narcolepsy type 1 over a 12-week period. Participants receiving twice-daily oveporexton experienced significant improvements in mean sleep latency, excessive daytime sleepiness, and the weekly cataplexy rate compared to those on a placebo. While adverse events such as increased urinary frequency and transient insomnia were more common in the treatment groups, most side effects were mild to moderate in severity. These findings confirm that addressing the underlying orexin deficiency successfully alleviates core narcolepsy symptoms and enhances overall quality of life.
